Head-to-head comparison

AOD-9604 vs HGH Fragment 176-191

Compare AOD-9604 and HGH Fragment 176-191 — structural differences, lipolytic mechanism, preclinical evidence, stability and the reasons researchers select one over the other for fat-metabolism studies.

Structural relationship

HGH Fragment 176-191 is the native C-terminal 16-amino-acid fragment of human growth hormone (residues 176 to 191). AOD-9604 is the same 16-residue fragment with an additional N-terminal tyrosine (Tyr-hGH 177-191), engineered by Metabolic Pharmaceuticals to improve stability and bioavailability during their Phase 2 obesity development programme.

Shared mechanism

Both peptides retain the lipolytic pharmacophore of HGH while lacking the IGF-1-mediated growth and insulin-resistance effects of full-length growth hormone. The proposed mechanism centres on stimulation of beta-3 adrenergic-receptor-mediated lipolysis in adipocytes and inhibition of acetyl-CoA carboxylase, biasing adipose tissue toward fatty-acid oxidation rather than lipogenesis.

Evidence base

AOD-9604 has the deeper human evidence file: Metabolic Pharmaceuticals advanced it through Phase 2b in adult obesity (Heffernan et al., 2001 and follow-up trials). The Phase 2 programme did not deliver the expected weight-loss magnitude over placebo to justify Phase 3 commitment, but the safety dossier is substantial. HGH Fragment 176-191 has not been advanced through a comparable human programme; the published literature is dominated by preclinical rodent lipolysis studies.

UK regulatory and research framing

Neither AOD-9604 nor HGH Fragment 176-191 is approved by the MHRA or EMA for human therapeutic use. Both are supplied as research peptides for laboratory and analytical use only. AOD-9604 has a stronger published safety dossier from its Phase 2 development, which is why some researchers prefer it as the reference compound for designing experiments that probe the lipolytic-fragment hypothesis.

Which to select

For most modern UK research the choice often comes down to the question being asked: AOD-9604 is preferred where a stabilised, well-characterised analogue with prior human safety data is required; native HGH Fragment 176-191 is preferred when the experiment specifically interrogates the unmodified endogenous C-terminal fragment. Both compounds remain substantially less-studied than the incretin class, and most researchers integrating them into broader metabolic protocols cross-reference the tesamorelin and CJC-1295 / ipamorelin literature for orthogonal mechanism comparison.
